Acetylcholinesterase (AChE) is a highly conserved vital enzyme responsible for hydrolysing acetylcholine, a stimulatory neurotransmitter. Inhibition of AChE results in a build-up of acetylcholine within muscle synaptic clefts, triggering prolonged muscle contraction often leading to fatal heart, lung and brain failure.

Tabun (GA) was the first nerve agent to be synthesized of the G-series organophosphates. Originally developed as insecticides by German chemist Gerhard Schrader in 1936, the human toxicity of these agents quickly became apparent, resulting in their stockpiling throughout World War two. GA would first see use during the Iran-Iraq war by Iraqi forces, highlighting the danger of these compounds resulting in a 1993 UN ban on their storage, manufacture and release.

Acetylcholineesterase (AChE) is an enzyme that terminates the cholinergic signalling molecule acetylcholine (ACh) through hydrolysis. ACh is a neurotransmitter that is released at neuromuscular junctions and brain synapses, which activates receptors and triggers muscle contraction. Accumulation of ACh leads to sustained contraction, paralysis, and potentially death from respiratory failure, which is why it is such a target for nerve agents. 

Aricept/E2020 is a medication for mild to severe Alzhiemer's disease. It works by being a highly selective and reversible inhibitor, limiting the function of AChE and therefore the breakdown of ACh. Aricept can improve symptoms such as memory loss and confusion in certain people, assisting those with Alzheimer's in their day-to-day life.

Acetylcholinesterase (AchE) is an enzyme that catalyze the hydrolysis of acetylcholine (the substrate/neurotransmitter) into acetate and choline and is present in animals and humans. AchE is a necessary enzyme that controls the levels and turns off the nerve signaling pathway that acetylcholine sends from the nerves to muscle cells resulting in the a muscle function. Failure of AchE to perform results in buildup of the neurotransmitter and may lead to continuous signals and end in paralysis or death. Tabun (GA) is an organophosphate nerve agent that binds covalently to the active site of the acetylcholinesterase. Tabun is therefore an inhibitor of acetylcholinesterase that inhibits the binding of prospecting acetylcholine molecules. Here, we will look at how Tabun binds with AchE active site and the complications surrounding it's reactivation.

Acetylcholinesterase (AChE) is an enzyme that terminates nerve signals by breaking down the neurotransmitter, acetylcholine (ACh), into acetic acid and choline. ACh is that is crucial for muscle movement and cognition. AChE ensures that the signal is brief and controlled for the target cells.

However, E2020, an Alzheimer’s Disease (AD) medication, works by slowing down AChE catalysis via non-covalent competitive inhibition. The lack of cholinergic activity is hypothesised to accelerate neurodegeneration by increasing tau tangle and amyloid plaques accumulation. By inhibiting the ACh breakdown, E2020 prolongs ACh activity in the synapses, enhancing target cell stimulation. The biomolecular model uses AChE of Tetronarce calfornica to depict the key interactions with E2020 which include hydrophobic interactions, π-π stacking, cation-π bonds, and hydrogen bonds.

Acetylcholinesterase (AChE) is the “cleanup crew” of the nervous system, rapidly breaking down acetylcholine to stop muscle signals and reset communication between nerve and muscle cells. This job happens inside a deep active-site gorge, where a catalytic triad drives fast hydrolysis.

Tabun, a highly toxic organophosphate nerve agent, acts like a fake worker that enters this site and shuts everything down. It forms a permanent covalent bond with the catalytic serine (Ser203), disabling the enzyme’s machinery. As a result, acetylcholine builds up, causing continuous muscle stimulation and disrupting vital functions such as breathing.

ACh is vital for signal transmission and muscle contraction, which makes breakdown of ACh incredibly important, as excess build up can cause response from nearby receptors.

Alzheimer’s disease (AD) is a disease that affects cognitive function, such as memory and thinking. AChE inhibitors decrease breakdown of ACh and leads to an increase in stimulation of muscarinic and nicotinic receptors (both of which are acetylcholine receptors), which helps with memory problems associated with AD. One of many medications used to treat AD is aricept (E2020), which reversibly binds AChE and has been found to downregulate neuroinflammatory responses, something that is found to have been associated with AD.
